Molecular mechanism of Clofazimine resistance in tuberculosis

چکیده
Drug resistance which is at present the main impendence to global tuberculosis (TB) control and drug- resistant (DR) activity of Mycobacterium in various strains have become major challenge worldwide. Nowadays, many researchers use different antibiotics for treatment DR-TB are often not well tolerated adequately efficient. A large group study from research published time described a regimen multidrug-resistant (MDR-TB) including Clofazimine (CFZ) as high incidence against MDR-TB; whereas therapeutic ways still limited, strategy repurpose existing anti- mycobacterial agents. CFZ one such drug that has recently devoted interest DR-TB. fat-soluble riminophenazine dye used TB, HIV, leprosy co-infected people shown action MDR TB now recommended by WHO treat agent with “unclear efficacy”. Although mode molecular mechanism yet entirely understood, it been exposed outer membrane its primary site, an extensive number mutant clofazimine found mutations rv0678 be most prominent respiratory chain ion transporters putative targets. This discussed comprehensive report, reactivity CFZ, provides new acuteness into clinical conduct this drug. Key words: Clofazimine, background, action, mechanism.

A STUDY OF MYCOBACTERIUM TUBERCULOSIS DRUG RESISTANCE IN PULMONARY TUBERCULOSIS
Tuberculosis remains a major public health problem in both developing and developed countries. Drug-resistant tuberculosis is an increasing health problem and serious challenge to tuberculosis (TB) control programs. Information about the susceptibility pattern of Mycobacterium tuberculosis isolates against anti-tuberculosis drugs is an important aspect to TB control. The objectives of the s...
متن کاملIdentification of novel mutations associated with clofazimine resistance in Mycobacterium tuberculosis.
OBJECTIVES Although clofazimine has been traditionally used to treat leprosy, there is recent interest in using clofazimine for the treatment of MDR-TB and drug-susceptible TB. However, the mechanisms of resistance to clofazimine are poorly understood. Here, we investigated the molecular basis of clofazimine resistance using resistant mutants isolated in vitro. METHODS We isolated 96 mutants ...

The antileprosy drug clofazimine is also of interest for the treatment of multidrug-resistant tuberculosis. To understand possible resistance mechanisms, clofazimine-resistant Mycobacterium tuberculosis mutants were isolated in vitro, and, unexpectedly, found to be cross-resistant to bedaquiline. Mutations in the transcriptional regulator Rv0678, with concomitant upregulation of the multisubstr...
متن کاملInsulin Resistance: Molecular Mechanism
Insulin resistance in skeletal muscle is present in humans with type 2 diabetes (non-insulin dependent diabetes mellitus) and obesity and in rodents with these disorders. Malonyl CoA is a regulator of carnitine palmitoyl transferase l (CAP I), the enzyme that controls the transfer of long chain fatty acyl CoA into mitochondria, where it is oxidized. In rat skeletal muscle, the formation of malo...
